Non-Canonical Function of Metabolic Enzyme in Tumor Progression

Metabolic reprogramming of tumor cells supports tumor initiation and progression. On one hand, metabolic enzymes provide materials and energy by altering metabolic pathways; on the other hand, they also exhibit non-metabolic functions. The non-canonical functions of metabolic enzymes play crucial roles in tumor progression. This review systematically summarizes key advances over the past decade, explaining novel mechanisms by which metabolic enzymes contribute to epigenetic regulation, signal transduction, and tumor microenvironment remodeling. Metabolic enzymes employ diverse mechanisms such as nuclear translocation, protein-protein interaction to directly modulate gene expression, signaling network rewiring, and immune responses. These findings provide a novel direc tion for developing precision oncology aimed at the non-canonical functions of metabolic enzymes.
