piRNA (PIWI-interacting RNA) is a kind of single strand non-coding RNA composed of 21-33 nucleotides, mainly expresses in germ cells, and plays a regulatory role by binding with PIWI protein. It shows that PIWI/piRNA can not only cleave and degrade mRNA directly in the cytoplasm, but also recruit a large number of epigenetic modified proteins to shut down gene expression by DNA and histone modification, so as to promote the safe and orderly development of gametes. This review summarizes the role of PIWI/piRNA mediated epigenetic regulation in reproductive development in order to promote the research and application of PIWI/piRNA in the field of life science in the future.
