Effects of the Supercritical Fluid Extract of Xiaoguishao on Telomerase Activity and Expression of Apoptosis-related Proteins in Human Gastric Cancer Cell Line

Abstract: To investigate the potential mechanism of apoptosis induced by the supercritical fluid extract of Xiaoguishao on human gastric cancer cell line BGC-823, PCR-ELISA assay was used to detect the telomerase activity and Western blot was used to analyze the expression of human telomerase reverse transcriptase (hTERT), p16, bax, and mutant p53 (mt p53). PCR-ELISA assay showed that in the extract of Xiaoguishao treating groups, the telomerase activity was significantly decreased compared with that in control group. Western blot results indicated that the expression of hTERT and mt p53 was decreased but the expression of p16 and bax was increased by the extract. The activation of p16 and bax and the suppression of telomerase activtiy and p53 might contribute to the apoptosis induced by the extract.
