Partial hepatectomy (PH), a basic protocol, is widely used to study the liver regeneration and tissue regeneration. This article introduced the experimental method of 2/3 PH in detail, used the C57 mice as the subject. In our study, survival rate of the mice after the operation could be to 90%. After PH, the mice liver and body weight ratio gradually increased, and 7~10 days after PH the liver weight can be increased to 90% of the normal liver weight. The hepatocytes stopped dividing after 10 days . The found of 2/3 PH is for us to lay the foundation of the future to study the mechanism of the liver regeneration.
