Expansion Microscopy and Its Application in Biomedicine

Abstract:

ExM (expansion microscopy) is a novel super-resolution imaging strategy based on the physical expansion of biological samples. It can bypass the resolution limit of traditional optical microscopes and achieve precise observation of cellular structures and macromolecular complexes at nanoscale. More importantly, in contrast to other optical super-resolution technologies, ExM is not restricted to one certain microscope. It has been successfully applied to diverse research objectives from cells to tissue, even to small biological individuals, showing the great value of ExM in biomedicine. This review will discuss the development, main types, research applications and future tendencies of ExM.
