Cholesterol homeostasis is essential for normal life activities of the organism. HMGCR (3-hydroxy-3-methyl glutaryl coenzyme A reductase)-mediated mevalonate pathway is a key step in the organism’s de novo synthesis of cholesterol. Therefore, the expression and regulation of HMGCR are important for the maintenance of cholesterol homeostasis. In the organism, the process of HMGCR from transcription to its reductase function is strictly regulated, including transcription, translation, protein stability and enzyme activity, and epigenetic regulation. This article comprehensively reviews and summarizes the research progress on the expression and regulation of this key enzyme, providing theoretical reference and ideas for the study of HMGCR regulating cholesterol metabolism.
