Effect of Xanthones from Gentianopsispaludosa on Mesenchymal Transformation of NCM460 Cells

This study investigated the effects of Xanthones from Gentianopsispaludosa on human NCM460 (normal colon mucosa cell) interstitial conversion. NCM460 cells were induced with 2.5 μg/mL LPS (lipopolysaccharide) for 14 d to establish interstitial transformation model, and were treated with GPX and PFD respectively. The expression of E-cadherin and Vimentin protein and mRNA were detected by IF (immunofluorescence), WB (Western blot) and RT-qPCR (real-time fluorescence quantitative transcription polymerase chain reaction), respectively. The results showed that compared with normal control group, the epithelial cell marker E-cadherin protein and mRNA in model control group were significantly down-regulated (P<0.01), interstitial cell marker Vimentin protein and mRNA were significantly up-regulated (P<0.01). Compared with the model group, the fluorescence intensity of E-cadherin was positively correlated with the dose of GPX, while the fluorescence intensity of Vimentin was decreased in a dose-dependent manner. The expression of E-cadherin protein and mRNA was up-regulated and that of Vimentin protein and mRNA was down-regulated in GPX medium and high dose groups (P<0.01), most obvious in high dose. The study showed that Xanthones from Gentianopsispaludosa could effectively inhibit colon EMT (epithelial mesenchymal transformation), which provided theoretical basis for further research and development.
