Regulation of Bone Marrow Microenvironment on Hematopoietic Stem Cells under Steady State and Aging

Bone marrow microenvironment was proved to regulate the self-renewal, proliferation, differentiation and migration of HSCs (hematopoietic stem cells) via different signal pathways and cytokines under steady state. Upon aging, the defective function of HSCs leads to the deterioration of hematopoietic integrity and changes of the age-related immune response, as well as increased susceptibility to anemia, autoimmune and myeloproliferative disorders. HSCs aging was initially thought to be induced by cell-intrinsic dysregulation. However, current studies are revealing that the bone marrow microenvironment not only regulates HSCs function under homeostasis, but also makes contributions to HSCs aging. This review will cover our understanding of HSCs regulation by bone marrow microenvironment under steady state and aging.
