Optimization of Transfection Condition of the EL Transfection Reagent in sophageal Squamous Cells

This study aimed to explore the influencing factor on the transfection effects of EL transfection eagent, which was used to transfect Hsa-miR-6743 plasmid into esophageal squamous cells. The esophageal squamous ells Eca-109, TE-1 and Eca-9706 were chosen as the research objects. Hsa-miR-6743 marked by GFP (green luorescent protein) was selected as a reporter gene. The fluorescence signals were detected by inverted fluorescence icroscopy to optimize the proportion of transfection reagent and plasmid. The results turned out that the transfection
efficiency of EL transfection reagent was influnced by cell types of esophageal squamous cells. The transfection fficiency of EL transfection reagent in Eca-109 cells was better than that in other two cells. The best detection ime-point of the optimal transfection efficiency was at 36 h in Eca-109 cells, and cell viability was not affected by L transfection reagent. The optimal transfection ratio of EL transfection reagent to Hsa-miR-6743 plasmid ranged rom 1:4 to 1:2. The transfection efficiency of EL transfection reagent was affected by cell types, the ratio of transfection reagent to plasmid, and transfection time.
