Molecular Mechanisms of Resistance to TRAIL-induced Apoptosis in Tumor

Abstract: Tumor necrosis factor (TNF)-related apoptosis-inducing ligand (TRAIL) is one of several members of the TNF gene super-family that selectively induces apoptosis of tumor cells, but not normal cells. However, several recent researches show that numbers of malignant tumors cells are resistant to apoptosis induced by TRAIL, and repeated application of TRAIL protein to some cancer cells that were originally sensitive to TRAIL-induced apoptosis results in acquired resistance. This review summarized the resistant mechanisms targeted the apoptosis pathway induced by TRAIL, and the molecular mechanisms of resistance mediated by other TRAIL activated pathway, such as Akt pathway.
