Abstract: V-raf murine sarcoma viral oncogene homolog B (BRAF) is a serine/threonine kinase with a key role in the mitogen-activated protein kinase (MAPK) pathway which regulates cell proliferation, differentiation and apoptosis. Several mutations have been observed in the gene coding area in a variety of human malignancies.Ameloblastoma is a benign but locally infiltrative odontogenic tumor, and a high frequency of BRAF mutations in ameloblastoma was reported in 2014, the highlight affair offer an expectation for future application of BRAFtargeted therapeutics. In this review we give an update on the clinical pathological and biological characteristics relevance of BRAF mutation and MAPK pathway in tumor, especially in ameloblastoma.
