β-lapachone Induced the Apoptosis of SW480 Colorectal Cancer Cell through Down-Regulation of the Peroxiredoxin V Expression

Abstract: Peroxiredoxin V (Prx V) is a thioredoxin peroxidase included in peroxiredoxins family, which is highly expressed in mitochondria, lysosomes and cytosols, and exhibits as scavengers for nitric oxide (NO) and reactive oxygen species (ROS). This study is aimed to explore the regulatory effect of Prx V on β-lapachone induced apoptosis of SW480 colorectal cell. To examine the SW480 cell viability, cellular ROS levels, apoptosis and the proteins levels, the MTT assay, fluorescence microscopy, flow cytometry and Western blot assay were performed. The results showed that β-lapachone treatments could decrease the Prx V protein level, which in turn increased the cellular ROS levels, then eventually activated the mitochondria signaling, and resulted in inducing the SW480 cell apoptosis. Our findings present a new therapeutic strategy for colorectal cancer.
