Nuclear Receptor TR3/Nur77 and Cancer Therapy

Abstract: Orphan nuclear receptor TR3 (also known as Nur77) is a product of an immediate-early gene encoded by NR4A1, and belongs to the steroid/thyroid/retinoid nuclear receptor superfamily. TR3 is widely involved in the regulation of biological processes, such as cell proliferation, differentiation, apoptosis and autophagy. It is considered as an important molecular target for anti-tumor drug design. TR3, as a transcription factor, regulates gene transcription and expression through targeting to the DNA response elements, and also functions as a regulator through interaction with other proteins in distinct subcellular localizations. Here, we review the functions and regulatory mechanisms of TR3 in tumorigenesis, and progress in the development of therapeutics with TR3 as the target.
