Abstract: The let-7 family, one of the most ancient and highly conserved microRNAs (miRNAs), plays critical roles in tumor suppressing. However, during normal development, accumulation of let-7 can be prevented by LIN28, a highly conserved RNA-binding protein, suggesting that over-expression of LIN28 might promote malignancy via repression of let-7. Emerging evidences suggest that LIN28/let-7 signal pathway can not only partly reveal the mechanism of tumorigenesis, but also guide the treatment of cancers.
