Abstract: Embryonic stem (ES) cells is one of the most popular topics in biomedical research since 1990s, and the newly developed RNA interference (RNAi) technology can silence the genes of interest quickly and efficiently, which would be a very useful tool to study the biological function of ES cells. Here, we review the mechanism of RNAi and the main methods to produce RNAi in ES cells, as well as the prospects of combining RNAi and ES cell manipulation for both basic research and future therapies.
