Abstract: To contrast the expression level of Xinjiang strain HPV-16 L1 and codon optimizing HPV-16 L1 in eukaryotic cells. Xinjiang strain HPV-16 L1 and codon optimizing HPV-16 L1 were cloned into pcDNA3.1 respectively. The recombinant plasmids were infused quickly into the mouse through tail vein or transfected into 293T cells, and then the transcripts of HPV-16 L1 were detected in the liver of the mouse and in 293T cells with RT-PCR, HPV-16 L1 protein was detected in 293T cells with Western blotting. The results show that Xinjiang strain HPV-16 L1 and codon optimizing HPV-16 L1 can be transcribed in the liver of the mouse and in 293T cells, and the L1 protein expression levels of Xinjiang strain HPV-16 L1 is lower than that of codon optimizing HPV-16 L1 markedly.
