The Effect of ANO1 on SCC-25 Oral Cancer Cell Line

Abstract: The specimens of 160 patients with oral squamous cell carcinoma were examined by immunohistochemistry. SCC-25 cell line was used to do many assays in vitro to find out how ANO1 affect on the migration of SCC-25 cells. ANO1 protein expression was significantly higher in oral squamous cell carcinoma than those in normal oral tissue, and showed significant correlation with clinical stage of oral squamous cell carcinoma and lymph node metastasis. The results of cell detachment assay, spreading assay, adhesion assay, boyden chamber migration assay and in vitro wound healing experiments showed that ANO1 overexpression favored the migration of SCC-25 cells; Niflumic Acid blocking chloride channel experiments showed that Niflumic Acid decreased the migration speed of SCC-25 cell. In summary, ANO1 may promote clinic metastasis of oral squamous cell carcinoma.ANO1 overexpression significantly increases the migration, invasion, spreading, detachment capacities of oral squamous cell carcinoma, and these biological characteristics are significant correlated with invasion and metastasis of the tumor cell. so ANO1 could become the new gene treatment target of oral squamous cell carcinoma.
