The Growth Inhibiting Effect of Oncolytic Adenovicus-Mediated TRAIL Combined with LiCl on Cancer Cells

Abstract: We investigated the killing effect of glycogen synthesis kinase 3 inhibitor LiCl combained with Ad.sp-E1A-E1B(Δ55 kDa)TRAIL-Flag (also known as Ad.sp-TRAIL-Flag) in vitro that oncolytic adenovirus carries on human tumor cells. MTT assay is used to detect inhibition on tumor cells proliferation by drug LiCl combined with tumor-specific virus Ad.sp-TRAIL-Flag, and combination effect on medication cytotoxicity are further detected by crystal violet experiment; Moreover, Western blot assays are performed to test the expression change of TRAIL. In addition, the apoptosis of these cancer cells was inspected by flow cytometry. The result shows that the growth inhibiting effect of LiCl combined with Ad.sp-TRAIL-Flag treatment on cancer cells is better than using them separately, so as to enhance the express of TRAIL through the signal path killing effects.
