Killing Effect to Hepatoma Cell SMMC-7721 by Dual-regulated Oncolytic Adenovirus-Mediated TSLC1 Combined with Adriamycin

Abstract: The study explores the combinational killing effects of dual-regulated oncolytic adenovirusmediated TSLC1 (Ad.sp-E1A(Δ24)-TSLC1) and adriamycin (ADM) against hepatoma cell SMMC-7721 in vitro. The method of RT-PCR was performed to test the expression of TSLC1 at the level of transcription. The morphological change was assessed by fluorescence microscope. MTT assay, crystal violet assay and Hoechst33342 staining were used to determine the growth inhibition effect and cytopathic effect of single or combination therapy on hepatoma cell SMMC-7721. The expression level of TSLC1, E1A protein was assayed by Western blot. These results demonstrated that combination therapy of Ad.sp-E1A(Δ24)-TSLC1 and ADM induces apoptosis characteristics more significantly than either single treatment, and low doses of ADM enhanced the killing effects of dual-regulated oncolytic adenovirus-mediated TSLC1.
