Effects of Quercetin on Growth of Colon Cancer Cell Line RKO

Abstract: MTT assay tested cell growth of colon cancer cell line RKO; cell cycle and cell apoptosis was observed by flow cytometry technique; RT-PCR and Western blot technique detected the expression of p21, p27 and p53. MTT assay showed that quercetin significantly inhibited cell growth and had dose-dependent effect; Flow cytometry technique showed that with 5 μmol/L quercetin the cell cycle was arrested in G0/G1 phase; apoptosis rates of 5, 10, 20 μmol/L groups were 23.4%, 24.2%, 47.9%, and apoptosis rate of the control group was 13.2%; mRNA and protein expression level of p21 and p27 increased, the expression level of pro-apoptotic protein p53 increased. Quercetin has significant growth inhibition on RKO cells. Cell cycle is arrested in G0/G1 phase by upregulating the expression of p21 and p27; quercetin may also induce apoptosis by upregulating the expression of p53.
