Effect of PXR Activation on Neuroblastoma Cells SH-SY5Y Apoptosis

Abstract: In this work we studied the specific antagonistic effect of PXR activation on neuroblastoma cell SH-SY5Y apoptosis induced by LPS. The expression level of PXR and CYP3A4 was measured by RT-PCR. Apoptosis of SH-SY5Y stained with Rhodamine 123, Annexin V-FITC/PI and growth phase of SH-SY5Y were detected by flow cytometry. The results showed that PXR activated by rifampycin significantly enhanced the expression levels of CYP3A4 and inhibited the apoptosis of SH-SY5Y cells. The analysis of cycle phase of cells indicated that rifampycin could arrest the cells at G0/G1 phase. The arresting efficiency of rifampycin decreased when PXR expression level was knockdown mediated with RNAi. PXR might inhibit the apoptosis of SH-SY5Y cells through arresting the cell cycle at G0/G1 phase.
