Establishment of A Poorly Differentiated Hepatocellular Carcinoma Cell Line TJ3ZX-01 and Research for Its Cellular Origin

Abstract: Cancer cell line TJ3ZX-01 were separated from poorly differentiated hepatocellular carcinomas specimen by the tissue dissection method, and their biological characteristics were identified. The cellular origin of the poorly differentiated HCC cell line was investigated with immunocytochemistry and flow cytometry analysis. Immunocytochemistry detection showed CK18+, CK19+, vimentin+, AFPlow, PCNA+ and Hep-, which demonstrated the cancer cell line had the characteristics of epithelial cells but not the obvious characteristics of liver cells for the markers of the liver cell were low. The liver cell and stem cells surface markers CD29, CD34, CD45, Thy-1 (CD90), CD133 and CD49f expressed in the cell line were stable with passages. A small subset of cells in the cancer cells migth has the characters of stem cells or progenitor cells, which indicated that this hepatocellular carcinomas cell line originated from stem cells or progenitor cells.
