Abstract: We reported here a new method for detection of radioactivity of radiolabeled nucleic acid probes. In this method, nylon filter and 0.4 mol/L of NaOH were used as chromatography media and chromatography solvent respectively. After chromatography, radiolabeled nucleic acid probes stayed at origin, while free nucleotides migrated to solvent front. Thus the radioactivity of radiolabeled nucleic acid and nucleotide could be measured respectively. This method is very simple, and could be finished in 30 min. On the other hand, this method leads to less contamination for the only radioactive waste was nylon filter itself after detection.
