The incidence and mortality of various tumors are still high, and the treatment of tumors is a major problem in clinical medicine. The development of tumor is inseparable from its “cradle”, that is, tumor microenvironment. Therefore, it is beneficial to explore the regulatory mechanism of the interaction between tumor and tumor microenvironment for tumor therapy. Studies have shown that STAT3 pathway plays an important role in the regulation of tumor microenvironment. This paper reviews the STAT3 pathway that activates CAFs (cancer associated fibroblasts), regulating the interaction between CAFs and tumor cells, CAFs and immune cells, and summarizes the tumor therapy drugs targeting STAT3 pathway.
