Effect of Quercetin on the Expression of TGF-β1, TNF in Alveolar Macrophage

Abstract: The silicotic alveolar macrophages were collected by bronchoalveolar lavage and were incubated in vitro. Alveolar macrophages were allocated into a blank group, a silicotic group and groups containing quercetin 10, 20, 40 μmol/L, respectively. Alveolar Macrophages were incubated for 2, 6, 12, 18, 24, 36 h. The expression levels of TGF-β1 and TNF in alveolar macrophage and P-p38MAPK in supernatant were analyzed. We found that the expression level of TGF-β1, TNF and P-p38MAPK were significantly (P<0.05) higher in silicotic groups than the blank groups in the same period. After treated with different concentration of quercetins, the expression levels of TGF-β1, TNF and P-p38MAPK were lower. The greater the concentration of quercetin, the less they were detected.We speculate that quercetin could down-regulate TGF-β1 and TNF expressions in AM upon silicotic exposure by regulating p38 MAPK signal transduction pathway.
