Abstract: The open field test (OFT) is a simple and reliable behavior test for rodents. Here we used it for the behavior analysis of GAT1 mutant mice to illustrate its basic principle and protocol. Each mouse was put into the open field and its behaviors were recorded for further analysis. We found that GAT1 knockout mice do not show any abnormalities in their locomotive activities, but they manifest lower levels of depression and anxietyrelated behaviors compared to their wild type littermates. These results suggest that GAT1 could potentially be used as a therapeutic target for the treatment of depression and anxiety.
