Mitosis in Primary Cultures of Silkworm

Abstract: The explants used for primary culture are tissue and semi-digestive tissue, which originate from the late embryonic tissue of silkworm (Bombyx mori). The optimal conditions for primary culture are Grace's medium supplemented with moderate amount of antibiotics, 20% fetal bovine serum and incubated at temperature 28 ℃, pH 6.4-6.8. The whole process of mitosis and abnormal mitosis such as abnormal cell division plate and endonuclear mitosis were found when tissue had been cultured more than 6 months. With the time of culture prolonged, divide ratio increased from 0.02±0.01 to 0.35±0.10, while the percentage of abnormal mitosis decreased from 8.0±1.6 to 3.2±1.0.
