A Simple Staining Method for Observation of Protoplast Nucleus and Protoplast Fusion in Rice

Abstract: A new fluorescent dye of Rhodamine B was screened. Protoplast nucleus staining with Rhodamine B became red under ordinary microscope or had red fluorescent light under fluorescent microscope. Staining by Rhodamine B or FDA (fluorescein diacetate), protoplasts from different plant can be distinguished because protoplast had red or blue fluorescent light respectively. Different types of protoplasts fusion can be clearly observed with this method.
